Gut microbiota from some participants in an earlier trial and an animal model

Were changes in behavioral ratings linked to changes in the gut microbiome?

Human follow-up analysis + animal experiment

Key findings

Communication changes in a small group of children

A pre–post change in the communication domain was reported for six children after 12 weeks.

Overall microbiome changes in humans remain unclear.

No clear change in the overall gut microbiome composition of the children was found; associations between some taxa and assessment changes were explored.

The mouse results came from a separate experiment.

In mice, social behavior and some microbial measures changed. The administration route also differed from the human study.

Detailed results

A study examining human data alongside mouse experiments

The human analysis included six boys with ASD aged 4–7 from Ou's earlier trial and 11 comparison children. The boys with ASD provided behavioral assessments and stool samples before and after 12 weeks of intake. The comparison children were used to compare gut microbiome features.

In a separate experiment, sulforaphane was injected intraperitoneally for 28 days into a mouse model of maternal immune activation during pregnancy. Social exploration of a novel mouse increased.

Associations were found between some communication changes and specific taxa.

Improvement was observed in half of six children, and the mean pre–post change in the OSU nonverbal communication subscore was significant. The researchers also analyzed correlations between changes in the relative abundance of certain microbial taxa and behavior.

Neither humans nor mice showed a significant increase in overall microbial diversity. The taxa that differed after intake were also not the same in the two species.

Correlation between gut taxa and behavioral changes

Correlations between behavior and microbial taxa were explored in six boys with ASD. This was a follow-up mechanistic analysis of an earlier clinical trial; factors such as diet could also affect the findings.

Study design

Some children's data and stool samples from Ou's earlier trial were analyzed. Separately, behavior and gut microbiota were studied in a maternal immune activation mouse model.

Participants
Six boys with ASD aged 4–7 from an earlier trial
11 comparison children · parallel mouse experiment
Intake and observation
Human: GLR and myrosinase tablets for 12 weeks
Mouse: intraperitoneal SFN injection for 28 days
Measures
OARS-4 communication and other assessments, 16S rRNA microbiome analysis, and mouse social behavior

Study context

A follow-up analysis of participants overlapping the Ou trial was combined with an injection experiment in mice. The paper belongs to a 2023 volume but was published online in January 2024.
