A multicenter trial of 108 participants and results by rater
Did caregiver and clinician ratings agree in the same trial?
Human · multicenter randomized double-blind controlled trial

Key findings
Results differed by rater.
In a multicenter trial of 108 participants, clinician ratings improved, and about one-third of participants had score reductions of at least 30%. Caregiver ratings showed no significant difference between groups.

Results also differed by rater in the multicenter trial of 108 participants.
A 12-week randomized double-blind trial in China assessed more children across multiple centers than earlier small studies. Caregiver scales showed no significant difference between sulforaphane and placebo, while clinician scales reported improvement in the sulforaphane group.
| Assessment | Reported results |
|---|---|
| Caregiver-completed scales | No significant difference versus placebo |
| Clinician-rated scale | Significant improvement in the sulforaphane group |
| Exploratory analysis by age | A larger effect was reported in participants older than 10 years. |

What does a “30% reduction” mean?
The authors reported that approximately one-third of participants had assessment scores at least 30% lower at 12 weeks. Improvement was seen in clinician ratings; caregiver ratings were not significant.
What additional analyses show
The authors reported effects across several intelligence levels, with a larger effect in participants older than 10 years. They also proposed differences between raters for follow-up research. Yang's study is a subsequent microbiome analysis of some participants in this trial.
Study design
Tablets and placebo were compared for 12 weeks across several centers. Caregivers and clinicians separately rated children's behavior and social responsiveness.
